I recently had the same problem. On your computer you need to get to a place that searches for all files and folders, then search itunes library, and right click on one go to properties, then uncheck read-only. do this to all of the results except for itunes music library. hit apply then OK for each of them. after that, launch itunes again. that worked for me on XP and should work on Vista too.

Hi guys I have some sort of a problem,,,
you see,, I have windows Vista,,, and I used to have Itunes 9 ((the one I used to upgrad my iphones 3gs and resulted in locking it- so I can’t use it unless for emergency calls ))
I’ve been waiting so long for Dev team to come up with a way to unlock the iphone 3gs and downgrade the baseband,,now i must have iTunes 8.2.1 to unlock my iphone ((iTunes 8.2 now supports iPhone or iPod touch with the iPhone 3.0 Software Update.Tunes 8.2 also includes many accessibility improvements and bug fixes. iTunes 8.2.1 provides a number of important bug fixes and addresses an issue with verification of Apple devices.))…
After I uninstalled the Itunes 9 from my computer ,, I downloaded the iTunes 8.2.1 and every time I want to open it a dialog box appears telling me ((the file “iTunes Library.itl” can’t be read because it was created by a newer version of iTunes ))
